高效液相色谱-串联质谱法同时测定表层水体中5类40种抗生素

建立了固相萃取-高效液相色谱-串联质谱（SPE-HPLC-MS/MS）同时检测表层水中5类40种抗生素的分析方法。水样经过滤、固相萃取柱富集净化后，以乙腈-0.2%（v/v）甲酸水溶液为流动相进行梯度洗脱，采用电喷雾电离源，在多反应监测、正离子模式进行定性定量分析。结果显示，40种抗生素在1~200 μg/L水平下线性关系良好，平均加标回收率为41.3%~112.6%。采用该方法对长江南京段表层水体进行检测，共检出13种抗生素，含量为13.4~780.5 ng/L，其中喹诺酮类抗生素恩诺沙星检出率达100%，大环内酯类抗生素克林霉素最高检出水平达739.4 ng/L。该法高效、灵敏、可靠，可用于实际水样中多种抗生素的分析。     

The Pharmacophore Hypothesis of Novel Inhibitors for Aurora A Kinase

A three-dimensional pharmacophore model was developed from a series of inhibitors of Aurora A kinase to discover new potent anti-cancer agents using the HypoGen module in the Catalyst software. The pharmacophore model was developed based on the structure of 20 currently available inhibitors, which were carefully selected from the literature. The best hypothesis （Hypo 1） was defined by four features： one hydrogen-bond donor and three hy- drophobic points, with the best correlation coefficient of 0.909, the lowest rms deviation of 1.563, and the highest cost difference of 99.075. The Hypo 1 was then validated by a test set consisting of 24 compounds and by a cross-validation of 95% confidence level through randomizing the data using the CatScramble program, which suggested that a predictive pharmacophore model had been successfully obtained.     

Resonance Rayleigh scattering method for the recognition and determination of double-stranded DNA using amikacin

In weakly acidic buffer medium, the interaction of amikacin with calf thymus DNA, yeast RNA and denatured DNA has been investigated by using resonance Rayleigh scattering (RRS) technique. The result shows that calf thymus DNA is capable of enhancing the RRS intensity of the amikacin, while yeast RNA and denatured DNA have very little enhancement effect. Based on the characteristics, a sensitive assay for detecting double-stranded DNA in the presence of denatured DNA and yeast RNA has been developed. The enhancement of the RRS signal is directly proportional to the concentration of double-stranded DNA in the range 0.02-12.0 μg ml⁻¹ for calf thymus DNA and its detection limit (3σ) is 2.5 ng ml⁻¹. The method shows a wide linear range and high sensitivity, and almost no interference can be observed from RNA, denatured DNA, amino acid and most of the metal ions. The trace amounts of nucleic acid in synthetic samples and practical samples are determined with satisfactory results. Therefore, the proposed method is promising for as an effect means for recognition in vivo and determination in situ of double-stranded DNA.     

A convenient method for the preparation of oxazaborolidine catalyst in situ using (S)-α,α-diphenylpyrrolidinemethanol,tetrabutylammonium borohydride,and methyl iodide for the asymmetric reduction of prochiral ketones

An oxazaborolidine catalyst is readily prepared in situ at 25 °C in THF using (S)-α,α-diphenylpyrrolidinemethanol and borane generated from tetrabutylammonium borohydride/CH₃I reagent system. The oxazaborolidine/BH₃ reagent system prepared in this way is useful for the reduction of prochiral ketones to the corresponding alcohols with up to 99% ee.     

毛细管区带电泳法测定复方甘草片中的甘草酸、甘草次酸、吗啡和苯甲酸钠

 在紫外检测波长为 2 2 8nm、电压为 14kV、背景电解质为 5 0mmol/L硼砂溶液、内标为氢氯噻嗪的条件下 ,用毛细管区带电泳法对复方甘草片中的甘草酸、甘草次酸、吗啡和苯甲酸钠进行了定量分析。结果甘草酸、甘草次酸、吗啡和苯甲酸钠的相对峰面积 (组分与内标的峰面积之比 )与各自的质量浓度之间呈良好的线性关系 ,上述 4种组分的平均回收率分别为 98 2 % ,97 3% ,97 1%和 97 5 %。方法简便、快速、准确。     

Diastereoselective synthesis of functionalized tetrahydro-γ-carbolines via a [3 + 3] cycloaddition of 2,2′-diester aziridines with β-(indol-2-yl)-α,β-unsaturated ketones

A Sc(OTf)₃-catalyzed [3 + 3] cycloaddition of 2,2′-diester aziridines with β-(indol-2-yl)-α,β-unsaturated ketones was developed, affording polysubstituted tetrahydro-γ-carbolines in single diastereoisomers in good to excellent yields.     

A new isocoumarin from bark of Pellodendron chinense

A new isocoumarin, 3-acetyl-3,4-dihydro-5,6-dimethoxy-1H-2-benzopyran-1-one, was isolated from the cortex of Phellodendron chinense schneid. The structure was determined on the basis of spectroscopic evidences.     

Radioimpurity identification by retention index in tritium labeling

Retention indexes (RI's) on SE-30 and Carbowax 20M columns are characteristic and can be used for identification purposes. A method for predicting RI on the basis of the number of atoms and contributions from substituents and functional groups is discussed. This method establishes a structure retention index relationship (SRIR), capable of relating structure to RI and is useful for suggesting structure to match with radioactive peaks. Examples of labeled side products tentatively identified in this manner are given.     

New bioactive triterpenoid from Oldenlandia cantonensis How

A new pentacyclic triterpenoid, urs-12-en-29α-oic acid-3β-ol (1), was obtained from the ethanol extract of Chinese herb Oldenlandia cantonensis How. The structure was elucidated by spectroscopy methods, including nuclear magnetic resonance (NMR) (1D and 2D), infrared spectroscopy (IR), and mass spectrometry (MS). 1 exhibited significant inhibitory activity against the human DNA topoisomerase I (hTopo I), the cancer cell lines BEL-7402 and MCG-803, with the IC₅₀ values 12.0, 6.5, and 8.0 μg/mL, respectively. The volatile oil, the fraction of petroleum ether: EtOAc = 20:1 (V/V) on Si gel chromatography, was also quantitatively analyzed by gas chromatography mass spectrometry (GC-MS). As a result, 60 compounds were identified. Among them, the long chain aliphatics, terpenes and steroids, as the representative structure type, were found with percentages of 36.16%, 6.42% and 9.28%, respectively. Rhodium-catalyzed CH activation/cyclization of enaminones with sulfoxonium ylides toward polysubstituted naphthalenes

A rhodium-catalyzed ortho-CH functionalization and annulation between enaminones and sulfoxonium ylides was developed, affording a series of multi-substituted naphthalenes in good to moderate yields with excellent functional group compatibility. The procedure featured with enaminone acting as both a directing and cyclization bifunctional group, and the application of sulfoxonium ylide in CH functionalization.
